Laboratory data were collected in 2021 on live fathead minnow sperm motilities by using computer assisted sperm motion analysis. Flow cytometry data were collected on the testicular cells; data are in standard format that allows files created by one type of acquisition hardware and software to be analyzed by any other type. This study on potential reproductive effects was performed in conjunction with colleagues working at a study site in Cape Cod, MA. This site has documented PFAS contamination due to firefighting compounds. The mobile laboratory was configured to perform continuous flow experiments with contaminated source water, reference water, and fish acclimation differences. Fathead minnows (Pimephales promelas) were exposed for either 0, 7, or 21 days in the various treatment groups. One testis per fish was shipped overnight to WARC Lafayette. To assess potential effects on male reproductive parameters, live sperm motilities were measured by computer assisted sperm motion analysis, and flow cytometric endpoints were collected, including sperm counts per tissue mass, and proportions of haploid and diploid forms were calculated from preserved samples. These data will be included with the larger study measuring multiple fish endpoints. Not all motility readings could be used because the commercial deliveries were late.
