Lipin2 limits macrophage proinflammatory response.

Alterations in Lpin2 encoding the phosphatidate phosphatase Lipin2 cause the autoinflammatory disorder Majeed syndrome. Lipin2 deficiency enhances lipopolysaccharide (LPS)-induced NLRP3 inflammasome formation in macrophages. To determine the physiological role of Lipin2 at regulating macrophage proinflammatory signaling, we knocked out Lpin2 with CRISPR/Cas9 in murine macrophage RAW-D cells and conducted a microarray expression analysis with LPS-treated or untreated WT and Lpin2 knockout (KO) cells.
