What genes does the transcription factor Blimp1 regulate in Tfr cells? Overall design: CD4+ T cells were isolated from the spleen from Wildtype (WT) and Blimp1-flox (Blimp1) Foxp3-cre mice 36 days after 2x oral challenge with peanut + cholera toxin. Cells were isolated via a CD4+ T isolation kit (Miltenyi Biotec). Foxp3+YFP+ CXCR5+ PD1+ cells were further isolated by FACS (FACS Aria). Total RNA was collected from freshly sorted TFR cells using the RNeasy Plus Micro kit (QIAGEN) and subjected to RNA-seq
