We used I-BET 151, an isoxazoloquinoline that specifically inhibits interaction of BET proteins with acetylated histones to restrict inflammatory tissue priming in a mouse model of iterated monosodium urate (MSU) crystal-induced arthritis. Systemic administration of I-BET 151 abolished the enhancement of arthritis upon repeated injection of MSU crystals. Differential gene expression analysis of cultured synovial fibroblasts derived from mouse joints 9 days after the first (non-primed) or the second (primed) injection of monosodium urate (MSU) crytsals with or without treatment with I-BET151 in vitro or in vivo. Three samples from each category (non-primed, primed, primed + treated in vivo, primed + treated in vitro) were used.
