This data were collected during Science week at Faculty of Nuclear Sciences and Physical Engineering, 2025 (https://tydenvedy.fjfi.cvut.cz) Crystallization phase diagram Experiment: Hanging drop vapour diffusion crystallization experiment. Gradient of salt concentration and protein crystalization Crystallization conditions: 50mM Na acetate pH 4.0, varying concentration of NaCl, reservoir volume 400ul. Drops 1ul+1ul and 1ul+2ul. Proteine concentrations 20-100 mg/ml File name: plate number-well-drop Columns: 1 - 20mg/ml; 2- 40mg/ml; 3 - 60mg/ml; 4 - 80 mg/ml; 5 - 100mg/ml Plate 1: Row A - 0 M NaCl; B - 0.5M NaCl; C - 1.0 M NaClPlate 2: Row A - 1.5 M NaCl; B - 2.0M NaCl; C - 2.5 M NaClPlate 3: Row A - 3.0 M NaCl; B - 3.5M NaCl; C - 4.0 M NaCl Drops: 1a, 1b - 1ul protein +1ul precipitant; 2a, 2b - 1ul protein + 2ul precipitant Images recorede after 24 hours at 20C using cross polarized light, Nextal 3x5 plates with ruber seal Diffraction experiment Diffraction data collected using D8 Venture (Bruker) with MetalJet D2+ and Photon III M28 at 100K. Crystallization conditions: 50mM Na acetate pH 4.0, 1M NaCl, 30% PEG 3350, reservoir volume: 400 ul, drop: 1ul+1ul, lysozyme at 50mg/ml. Flash frozen in liquid nitrogen, no cryprotection. Crystal size about 80um Exposure time 3s and 10s, oscilation 0.3° per frame. 20 runs with various goniometer settings.
