Efficacy and specifcity of novel photoreceptor-specific promoters were evaluated in wild-type and mutant (PDE6Ã-RCD1 and RPGR-XLPRA2) dogs at advnced stages of retinal degeneration using massively parallel reporter assays. Overall design: Each dog was injected subretinally with a pool of AAV5-promoter-GFP vectors. Five weeks post-injection, neuroretinal punches were collected and dissociated into single cells for scRNASeq analysis.
