We screened for potential warfarin dose-associated CpGs by comparison of genome-wide DNA methylation between the case groups(low dose group and high dose group) and control groups(medium dose group).We isolated DNA from peripheral blood cell samples by using the QIAamp DNA Blood Mini Kit (Cat.#51306; Qiagen, German). DNA bisulfite conversion was done in a bisulfite batch with the Zymo EZ DNA MethylationTM Kit (Cat.#D5001, Zymo, USA). Genome-wide DNA methylation was measured by Illumina Infinium HumanMethylation 450 K BeadChip (Cat.#WG-314-1001, Illumina, USA).Data from genome-wide methylation were analyzed using the GenomeStudio Software (version V2011.1, Illumina, San Diego, CA, USA).我们通过对比病例组（低剂量组与高剂量组）与对照组（中剂量组）的全基因组DNA甲基化水平，筛选出与华法林剂量相关的潜在CpG位点（CpG）。本研究采用德国Qiagen公司生产的QIAamp DNA血液微量试剂盒（货号：51306）从外周血细胞样本中提取基因组DNA；使用美国Zymo公司生产的Zymo EZ DNA甲基化试剂盒（货号：D5001）进行批量亚硫酸氢盐转化；采用美国Illumina公司生产的Illumina Infinium人类甲基化450K芯片（货号：WG-314-1001）检测全基因组DNA甲基化水平；并使用美国加利福尼亚州圣地亚哥市Illumina公司开发的GenomeStudio软件（V2011.1版本）对全基因组甲基化数据进行分析。
