454 Sequencing of transcriptome libraries representing the 3'-ends of cDNAs was used to analyze the transcriptomes of nodules and roots of the actinorhizal plant Datisca glomerata. The transcriptome comparison was used to analyze functions relevant for nodule development, structure, metabolism, and the stable internal accommodation of Frankia via the identification of genes differentially expressed in nodules compared to roots. The sequencing effort resulted in 6708 unique contigs of which 1128 (ca. 17 %) could be assigned a homology to a protein encoded by another organism, or a gene previously characterized from D. glomerata, by running blastx. We could establish that 254 sequences were significantly up-regulated in either roots or nodules. Among the sequenced contigs we found among many interesting transcripts a D. glomerata homolog to the A. glutinosa dicarboxylate tranporter AgDCAT1.
