Bulk RNA-seq analysis was performed across a 72 hr timecourse for JURL-MK1 cells in which the dopamine receptors DRD3 or DRD4 were knocked down by lentiviral mediated shRNAs in vitro. Knockdown cells were also treated with and without a TKI (nilotinib. Overall design: DRD3 was knocked down in JURL-MK1 cells using two different shRNA constructs (sh46 and sh47). DRD4 was knocked down in JURL-MK1 cells using two different shRNA constructs (sh53 and sh55). Controls were shRNA for scrambled. Post-transduction of JURL-MK1 cells with shRNAs, total cellular RNA was harvested on days 1,2,3 of time course experiments for each knockdown construct. sh53 DRD4 knockdown cells were also +/- tyrosine kinase inhibitor (TKI) nilotinib (2 nM) for 2 or 3 days. Independent biological replicate experiments were performed in duplicate (n=2) or triplicate (n=3) for each knockdown condition and each timepoint.
