In the present study we created and analyzed endothelial cells from two separate iPSC clones from the PBMCs of three different normal individuals using footprint-free Sendai virus, under highly coontroled differentiation and treatment Endothelial differentiation of iPSCs grown in EGM2 media, using RPMI-B27 medium with 6uM CHIR induction for 2 days (d0-d2) and 2 uM CHIR treatment for two days (d3-d5). iPSC differentiated endothelial cells (ECs) were treated with simvastatin (1uM ) for 72 hrs. For diabetic modeling, iPSC-ECs were treated with 25mM glucose and 20 nM insulin and for 1 week.
