Radial glial cell signatures with FGFR3 hypomethylation and overexpression characterize central neurocytoma

We explored the genomic events behind the rare neoplasms of the central nervous system, central neurocytoma (CN) with multi-omics approach including whole-exome sequencing, bulk and single nuclei RNA sequencing, and methylation sequencing. We found FGFR3 hypomethylation leading to its overexpression as a major event in the ontogeny of CN which affected crucial downstream events like aberrant activity of PI3K-AKT and neuronal development pathways. Furthermore, we showed the similarities of CN with radial glial cells by gene markers and CN tumor cells and postulated the tumorigenesis of CN is due to dysregulation in the differentiation process from radial glial cells to neurons. Our data exhibited the role of FGFR3 as one of the leading drivers in the tumorigenesis of CN.
