RNA-sequencing analysis of control and SMYD3-knockdown MDA-MB-231 tumors from the mammary glands of orthotopic xenograft NOD/SCID mice. SMYD3 (also known as KMT3E), a hisone H3 lysine K4 methyltransferase, is highly expressed in several human cancers, including colorectal and breast carcinomas. Results provide insight into the transcriptional regulation of SMYD3 in breast cancer. Overall design: MDA-MB-231 cells stably infected with lentiviruses encoding either control (pGIPZ vectror) or SMYD3 shRNA (pGIPZ-shSMYD3) were orthotopically injected into mice. Total RNA was isolated from control and SMYD3 knockdown MDA-MB-231 tumors from the mammary glands of orthotopic xenograft NOD/SCID mice using Trizol reagent. For each of the 2 conditions, 2 biological replicates were included. In total, 4 RNA-sequencin samples were analyzed; 2 controls and 2 SMYD3 knockdown MDA-MB-231 tumors.
