This study was designed to investigate the specificity of dendritic cells migrated from the tumor site to the thymus. Overall design: Mice were inoculated subcutaneously with tumor cells and maintained for 12 days. Subsequently, Yellow-green microspheres (YG) was administered by intratumoral injection. Two days after YG treatment, thymic dendritic cell subsets were isolated for downstream analysis. Thymic plasmacytoid dendritic cells (pDCs) and Sirpa? conventional dendritic cells (Sirpa? DCs) were sorted using a BD FACSAria⢠III cell sorter. pDCs were defined as CD11c^low B220? cells, while Sirpa? DCs were identified as CD11c^high Sirpa? cells. The following samples were included in this study: YG+ pDC samples: thymic YG+ pDCs isolated from tumor-bearing mice with YG injection PBS pDC samples: thymic pDCs isolated from PBS-treated mice PBS Sirpa+ DC samples: thymic Sirpa+ DCs isolated from PBS-treated mice Total RNA from sorted cell populations was extracted using TRIzol reagent (Invitrogen) according to the manufacturer's instructions and used for downstream transcriptomic analysis.
