mRNA expression profiles of teratomas derived from mouse embryonic stem cells containing Sox2 WT and Sox2 T258A mutations Overall design: The T258A mutation in which threonine, the 258th residue of Sox2 was mutated to alanine, was introduced into one allele of E14 cells using the CRISPR-Cas9 editing system. E14 wild-type and Sox2 T258A mutant cells were transplanted into nude mice. Whole transcripts of resulting teratomas were analyzed by RNA-seq.
