Fundulus heteroclitus scales were chemically fixed with 2.5% glutaraldehyde, 2% formaldehyde in 0.1M cacodylate buffer (pH 7.3), then post-fixed in 4% osmium tetroxide and stained en bloc in 1% uranyl acetate. The scales were then dehydrated in a graded series of ethanol and infiltrated with Spurr's resin. Thin sections of 70 nm were trimmed using a diamond knife and post-stained in uranyl acetate and lead citrate. This micrograph was imaged using a Phillips CM 100 transmission electron microscope at an accelerating voltage of 80 kV.以2.5%戊二醛、2%甲醛溶于0.1M二甲胂酸缓冲液（cacodylate buffer，pH 7.3）中对底鳉（Fundulus heteroclitus）鳞片进行化学固定，随后用4%四氧化锇（osmium tetroxide）进行后固定，并以1%乙酸铀酰（uranyl acetate）进行整块染色。随后将鳞片经梯度乙醇系列脱水，并用斯普尔树脂（Spurr's resin）进行渗透包埋。使用金刚石刀修块后切取70nm超薄切片，再经乙酸铀酰与柠檬酸铅（lead citrate）复染。该显微图像采用飞利浦CM 100透射电子显微镜（transmission electron microscope），以80kV加速电压采集获得。
