The dataset provides detailed information on DNA damage induced by X-rays in glioblastoma multiforme (GBM) cell lines, which exhibit varying levels of radiosensitivity. The data were collected from fluorescence microscopy analysis of the yH2AX assay, a widely used marker of DNA double-strand breaks.The dataset includes:Raw data documenting the number and area of yH2AX foci per cell nucleus, measured in three different GBM cell lines (G44, G88, U251) after exposure to three radiation doses (2, 4, 6 Gy) and an unirradiated control. Additionally, it contains time-course data from the U251 cell line showing the kinetics of foci disappearance at six time points post-irradiation (0.5, 1, 2, 4, 6, and 24 hours). This data is organized in seven separate sheets within the file "Raw data yH2AX assay.csv".Compressed images from the experiments, captured using the Olympus IX83 Cellvivo microscope and processed with cellSens software. These images are organized into folders based on the cell line (G44, G88, U251) and radiation dose, showing visible yH2AX foci as a marker of DNA damage. One set of folders illustrates radiosensitivity, while another set documents the kinetics of DNA damage repair in the U251 cell line at different time points after irradiation. The images are available in two zipped files: "Radiosensitivity_merged.zip" and "kinetics.zip".ImageJ macro script ("macro_foci_counting.txt") designed to quantify DNA damage by counting the number of yH2AX foci in the cell nuclei from the images. This script is compatible with ImageJ version 1.53t and automates the analysis process to ensure consistent and accurate quantification of post-irradiation DNA damage.
