We report the derivation of two distinct stem cells of the trophectoderm lineage from human pluripotent stem cells. The first is a CDX2- stem cell equivalent to primary hTSCs – they both exhibit identical expression of key markers, are maintained in culture and differentiate under similar conditions, and share high transcriptome similarity. The second is a CDX2+ putative human trophectoderm stem cell (hTESC) with distinct cell culture requirements and differences in gene expression and differentiation relative to hTSCs. RNA sequencing was conducted on the following cells/conditions: two primary hTSC lines (CT29, CT30), undifferentiated hESCs (two cell lines - H1, H9), hESCs after 3 days of differentiation to trophoblast (H1-day3, H9-day3), hTSCs derived from hESCs (H1-hTSC, H9-hTSC), hTESCs derived from hESCs (H1-hTESC, H9-hTESC). Four biological replicates were used for each cell type/condition (10x4=40 samples total)
