Ankrd22 knockout mice were commissioned for construction by Cyagen Biosciences using CRISPR/Cas9 technology. The F4/80+ cell subset, isolated from mouse peritoneal cells through FACS sorting, corresponds to the population of mouse peritoneal macrophages. Single-cell suspension was added to the 10x Chromium chip according to the instructions for the 10x Genomics Single Cell 3 v3 Reagent Kit, with the expectation of capturing 8000 cells. cDNA amplification and library construction were performed according to standard protocols. Libraries were sequenced by LC-BioTechnology on an Illumina NovaSeq 6000 sequencing system at a minimum depth of 20,000 reads per cell. The animal experiments were approved by the Clinical Medical Ethics Committee of the Second Affiliated Hospital of Zhejiang University School of Medicine and the Ethics Committee of the Zhejiang Chinese Medical University Animal Experimental Research Center.
