Using sugarcane and rice as models to identify stress response transcripts in the orphan crop Fonio millet (Digitaria exilis). Transcripts were initially assembled using protein baiting and SRA datasets. These transcripts were used to design magnetic bead capture primers and transcripts were isolated from total RNA. These were converted to cDNA with the Oxford Nanopore cDNA sequencing kit and sequenced with ONT MinION prior to assembly with Canu.
