Step Notes. 1. Dispensed into white solid-bottom 1536-well MB Greiner plates with a Flying Reagent Dispenser (FRD). Reagent is kept on ice. Final concentration is 10 nM PI5P4Kα, 75 µM PI5P and 150 µM DPPS. 2. Plates centrifuged at 300×g for 10 seconds. 3. DMSO compound solutions transferred with a Kalypsys pintool. 4. Dispensed with a FRD. Final concentration is 5 µM ATP. 5. Plates incubated at room temperature. 6. Dispensed with a FRD. 7. Plates incubated at room temperature. 8. Dispensed with a FRD. 9. Plates incubated at room temperature. 10. Luminescence was measured with a ViewLux CCD Imager with a 20 second exposure time.
