Reduced representation library sequencing for Pacific white shrimp

We selected eight animals, and equal amounts of DNA (20*g in total) was pooled and submitted for complete digestion (18 hours) using the restriction endonuclease HaeIII (GG?CC) (New England Biolabs, Beverly, MA, USA). Digested genomic DNA was loaded onto a 0.9% agarose gel and fragments of sizes roughly 1-4 kb were cut and purified QIAEX II Gel Extraction Kit (Qiagen). Using a NanoDrop 1000 spectrophotometer, the final product was quantified to be 10.8 ng/*l with a total volume 50 *l. The massive parallel DNA sequencing was performed on an Illumina/Solexa Genome Analyzer II. We selected the option of single read and 75 cycles of sequencing on six lanes.
