Stability of copy number amplifications in Saccharomyces cerevisiae in the absence of selection

This project studies the stability of gene copy number variants (CNVs) in Saccharomyces cerevisiae in absence of the selection pressure under which they arose. Experimental evolution of budding yeast in nutrient-limited chemostats is known to recurrently select for amplifications of nutrient transporter genes. To investigate the fate of these CNVs upon return to the pre-selection environment, we evolved 15 unique CNV lineages (11 segmental and 4 whole-chromosomal amplifications) in nutrient-rich media: 3 strains for 110 generations, and the other 12 strains for 220 generations. Clones were isolated post-evolution from the 5 CNV lineages which lost their amplifications and reverted to one-copy. Here we have whole-genome sequencing data from all 71 strains- 15 CNV lineages and 56 revertant clones.
