Single-cell gene expression of circulating 41BBzeta CART19 cells isolated from lymphoma-bearing NSG mice treated with daily oral gavage of water or beta-hydroxybutyrate. Cells were captured and processed using 10X 3' GEX Reagent Kit v2 Overall design: Single cell RNA sequencing of circulating CAR T cells. In vivo design for single cell RNA-sequencing (scRNA-seq) on circulating CART19 cells: First, NSG mice were inoculated subcutaneously with 5e6 CD19? OCI-Ly18 cells and gavaged daily with oral beta-hydroxybutyrate, or. BHB (80mg per mouse, Ketone-IQ from HVMN) or vehicle (water). On day 0 post-implantation, mice received a curative dose of 5e6 human CART19 cells. Peripheral blood T cells were purified on day 17 following infusion, (n = 7 mice per arm), pooled amongst groups, and prepared for 3' scRNA-seq (10X Genomics).
