RIP1 inhibition protects TMJ from stress-induced TMJ OA

A compressive mechanical force-induced-TMJ OA rat model was used in the study. Lenti-virus targeting RIP1 was constructed in vitro and injected into the TMJ. Hematoxylin and eosin staining, TUNEL staining and micro-computed tomography were used to assess histological and morphological changes in the mandibular condyle. Immunohistochemistry, Western Blot and real-time PCR were performed to determine the changes of inflammatory, apoptotic and necroptotic markers.
本研究采用机械压迫力诱导的颞下颌关节骨关节炎(TMJ OA)大鼠模型。体外构建靶向受体相互作用蛋白1(RIP1)的慢病毒(Lenti-virus),并将其注射至颞下颌关节。通过苏木精-伊红染色(Hematoxylin and Eosin Staining)、TUNEL染色(TUNEL staining)与微型计算机断层扫描(micro-computed tomography),评估下颌髁突的组织学与形态学变化。采用免疫组织化学染色(Immunohistochemistry)、蛋白质免疫印迹(Western Blot)及实时荧光定量聚合酶链式反应(real-time PCR),检测炎症、凋亡及坏死性凋亡标志物的表达变化。
