The mechanism of XYP-001 in treating pulmonary fibrosis

We performed single-cell sequencing on the mice lung tissues of bleomycin induced pulmonary fibrosis model group and XYP-001 (Itraconazole) treatment group. Overall design: ICR female mice (22 – 25 g) were obtained from Shanghai Jihui Experimental Animal Breeding Co., Ltd (certificate: SCXK 2022-0009), divided into three groups: Control group (n = 12, did nothing), Model group (n = 12, 25 µL of 1 mg/mL bleomycin sulfate solution diluted with PBS (9041-93-4, Aladdin, China) was administered into the mice lungs by spray on day 0), XYP-001 treatment group (n = 12, model with 2.5 mg/mL XYP-001 (20100302, Joincare Haibin Pharmaceutical Co., Ltd., China) nebulized inhalation administration for 1 hour every day from day 0 to day 20). On day 21 lung tissues of mice were collected (left upper lobe of lung) and stored on ice in tissue preservation solution (DMEM:fetal bovine serum=9:1). Mix 6 animals each group (model group and XYP-001 treatment group) into one sample and send it for scRNAseq (10× Genomics) within 1 hour.
