The study synthesized and expressed 40 genes for each of two proteins (a DNA polymerase and a single-chain antibody) to investigate the impact of synonymous codon usage on protein expression levels in E. coli. The data, derived from measured protein yields and gene sequence characteristics, revealed strong correlations between codon bias and expression levels. Using partial least squares regression analysis, the researchers identified critical codons influencing expression and validated their findings by designing and testing new gene variants with optimized codon usage.
