In this experiment, we investigated the role of AQP4 and AQP10 in the low salt stress of razor clams. Firstly, the spatio-temporal pattern of AQPs distribution was obtained by measuring the expression of AQPs in various tissues of razor clam and gill tissues under low salt stress. Subsequently, dsRNA interference was used to knock down AQPs, and the heart rate, survival, and apoptosis rate of razor clams were measured to verify the role played by AQPs in osmoregulation of razor clams
本实验探究了水通道蛋白4(AQP4)与水通道蛋白10(AQP10)在缢蛏低盐胁迫中的作用。首先,通过检测缢蛏各组织以及低盐胁迫下鳃组织中水通道蛋白(AQPs)的表达水平,明确了水通道蛋白的时空分布模式。随后,采用双链RNA(dsRNA)干扰技术敲低水通道蛋白的表达,并检测缢蛏的心率、存活率与细胞凋亡率,以验证水通道蛋白在缢蛏渗透压调节中所发挥的功能。
