m6A-seq in NOMO-1, MA9.3RAS and MA9.3ITD cells with or without R-2HG treatment. Overall design: In sensitive leukemia cells we identified, R-2HG dramatically increases global N6-methyladenosine (m6A) modification via directly targeting and inhibiting FTO. We performed m6A-seq in NOMO-1 (one of the most sensitive cells) with or without R-2HG treatment for 48 hours to confirm whether R-2HG can dramatically up-regulates m6A modification and to determine which transcripts are with enriched m6A abundance after R-2HG treatment. We also performed m6A-seq in R-2HG- and PBS-treated sensitive cells (MA9.3ITD) with or without FTO knockdown, as well as R-2HG- and PBS-treated resistant cells (MA9.3RAS) with or without FTO expression to further analyze the effects of R-2HG and FTO on m6A modification.
