Single-cell RNA-seq of healthy human bone marrow. Overall design: Cryopreserved primary human bone marrow mononuclear cells were obtained from STEMCELL Technologies (catalog #70001) or from StemExpress (catalog #BMMNC025C). Frozen human bone marrow samples were thawed and dead cells were depleted using a dead cell removal kit (Miltenyi Biotec, 130-090-101). Cells were tagged with cell hashing (Stoeckius et al., 2018) oligo-tagged antibodies (BioLegend) according to manufacturer's instructions and pooled on a 10x Chromium instrument (10x Genomics) to generate barcoded single-cell beads-in emulsion (GEM) according to the manufacturer's protocol.
