Surface water microbe metagenome sequences (DP1.20281.001)

Surface water metagenomic samples are collected once per year during the approximate period of peak productivity. The metagenomics sampling occurs at the same time and location as surface water chemistry sampling. In wadeable streams, samples are collected near the downstream S2 sensor location. In lakes and rivers, samples are collected near the buoy sensor set at 1 sampling depth in unstratified systems and up to 3 sampling depth(s)in stratified lakes. Surface water grab samples are filtered on 0.22 um Sterivex capsule filters, capped and flash-frozen in the field. The filters are stored on dry ice in the field, and later are shipped frozen to an analytical facility for DNA extraction, sample preparation and shotgun metagenomic sequencing. Laboratory data are then delivered to NEON, where they undergo QC testing and are published on the NEON Data Portal.地表水宏基因组样本（surface water metagenomic samples）每年于大致的生产力峰值时段采集一次。宏基因组采样与地表水化学采样在同一时间与地点同步开展。在可涉水溪流中，样本采集于下游S2传感器（S2 sensor）附近区域；在湖泊与河流中，样本采集于浮标传感器（buoy sensor）附近：非分层水体设置1个采样深度，分层湖泊则最多设置3个采样深度。地表水瞬时水样通过0.22微米Sterivex胶囊式滤器（0.22 um Sterivex capsule filters）过滤后加盖，并在野外环境中快速冷冻。滤膜在野外阶段保存于干冰中，后续以冷冻状态运送至分析实验室，用于DNA提取、样本制备及鸟枪法宏基因组测序（shotgun metagenomic sequencing）。实验室生成的数据随后提交至NEON，经质量控制（QC）测试后，于NEON数据门户（NEON Data Portal）发布。
