Whole genome sequencing checks of WT (strain 8103) and four mutants from the NeMeSys complete collection of mutants in Neisseria meningitidis

To address one of the most important challenges in biology – determining the function of genes of unknown function – we constructed a complete collection of defined mutants in the important human pathogen Neisseria meningitidis. This collection named NeMeSys consists of individual mutants in 1,584 non-essential genes of strain 8013. As a quality control, Illumina whole genome sequencing was performed on the WT strain, which genome sequence was determined by Sanger sequencing more than 12 years ago, and four mutants in NMV_1687 (lnt), NMV_1812 (tatB), NMV_1934 (secB),and NMV_2236 (nlaB). WGS confirmed both the accuracy of the original genome sequence and that all the mutants contained the expected mutations.
