FOXO1 knockdown is toxic for BL cell lines.

We found that FOXO1 knock-down inhibits cell cycle progression in BL cell lines BL41, Namalwa and Jiyoye. Overall design: BL cell lines BL41, Namalwa and Jiyoye were transduced with sh_hFOXO1.H-10381 in pRSI12-U6-sh-UbiC-TagRFP-2A-Puro lentiviral vector (DECIPHER shRNA Library, Human Module I (Cellecta, https://www.cellecta.com/) obtained from BioCat, Heidelberg, Germany). On Day 4 after transduction total RNA was isolated with RNeasy mini kit (QIAGEN). RNA-sequencing as well as data processing were done by BGI (Hong-Kong). RNA libraries were prepared for sequencing using standard BGI protocols. SOAPnuke software was used for data filtering and Bowtie2 was used to map clean reads to reference. RSEM was used to calculate gene expression level and DEseq2 was used to detect DEGs. The human reference genome hg38 was used.
