CRISPR knock-out screen for NLRP3 R260W inflammasome-mediated cell death

The NLRP3 R260W mutation is associated with the pediatric autoinflammatory disease CAPS (Cryopyrin associated syndromes). NLRP3 R260W encodes constitutive active NLRP3 protein that induced inflammasome activation and inflammation without need for activation signal. Inflammasome activation leads to release of cytokines and inflammatory cell death (pyroptosis). This screen aimed to identify positive regulators of the inflammasome pathway induced by constitutive active NLRP3.
