Transcriptomes of rachilla tissues from two contrast groups differing for rachilla hair length were developed. The genotypes in these contrast groups include, the long rachilla hair types â Barke, Bowman (long) and their respective short rachilla hair counterparts Morex and BW-NIL-srh1. The rachilla tissues from the respective genotypes were collected from the central spikelets at rachilla hair initiation (W8.0), and rachilla hair elongation W9.5) stages. Total RNA from individual tissues was extracted using TRIzol reagent (Invitrogen) followed by 2-propanol precipitation. Genomic DNA residues were removed with DNase I (NEB, M0303L). High-throughput paired-end sequencing was conducted at Novogene Co., Ltd (Cambridge, UK) with Illumina NovaSeq 6000 PE150 Platform.
