A wild emmer wheat (Triticum dicoccoides accession 'Zavitan') genome assembly was constructed using a whole genome shotgun approach. With a target sequence coverage of 176x (based on an estimated genome size of 12 Gb), paired-end (450 bp) reads were assembled into contigs (L50 = 59 kb), which were then assembled into scaffolds (L50= 7 Mb, gap= 1.7%) using mate-pair reads (0.8, 2-4, 5-7, 8-10 kb) via the denovoMagic algorithm (NRGene ltd). Scaffolds were anchored to a high-density genetic map (>900,000 markers) to validate their integrity and then were assembled into super-scaffolds using Hi-C data (6.5 million connections). These analyses enabled to connect most of the scaffolds (10.1 Gb in total) and uniquely orient their physical positions within the 14 chromosomes of wild emmer wheat. Genome annotation was accomplished using RNAseq data from 20 libraries representing different tissues and time points. In total, 61,821 genes were identified with an average locus size of 5,994 bp.
