The anti-trimethyl-histone H3 (Lys27) (Millipore #07-449) antibody was used for the experiment. ~1000-2000 mosquito eggs at 15-18 hours after oviposition were used to prepare ChIP-seq library. Embryos were bleached in 50% bleach, homogenized and fixed in 1.8% formaldehyde/buffer A. 8-10 cycles of 10/10sec ON/OFF in lysis buffer with SDS and NP40 were performed on Bioruptor Diagenode machine for chromatin sonication. Chromatin immunoprecipitation was performed as previously described by Gomez-Diaz et al. 2014 using Agarose beads (ThermoFisher, 20421). ChIP-seq libraries were prepared using NEBNextUltra II DNA Library Prep Kit for Illumina (NEB, E7645).
