To understand the cellular composition and transcriptional phenotype of different cell populations in the mouse bleomycin induced fibrosis model, we performed RNAseq analysis on leukocyte, endothelial, epithelial and stroma (other) cell populations isolated from mouse lung samples. Overall design: Mice recieved intratracheal injection of either saline or bleomycin. We selected time point day 14, which was the peak of gene expression of most pro-fibrotic genes in this model. We isolated four different cell types from C57/B6 mouse lungs by FACS sorting: epithelial (EpCAM+), leukocytes (CD45+), endothelial (CD31+), and others (triple negative; EPCAM-CD45-CD31-). RNA was harvested via RNeasy kit (Qiagen). There were two groups, G1:C57/B6, male mice I.T. with Saline (n=5) ; and G2: C57/B6, male mice I.T. with Bleomycin (n=8).
