Effect of the SYK Inhibitor Entospletinib on Renal Inflammation and the Transition from Acute Kidney Injury to Chronic Kidney Disease Induced by Ischemia-Reperfusion Injury

We used single-cell RNA sequencing with the Evercode™ WT kit (Parse Biosciences) to profile kidney immune cells in a mouse model of ischemia-reperfusion injury (IRI), with or without treatment with the SYK inhibitor Entospletinib. The dataset captures immune dynamics at early and late stages of injury and was used to identify leukocyte populations associated with inflammation and its resolution, as well as the immunomodulatory effects of SYK inhibition in vivo. Overall design: Kidneys were collected from C57BL/6 mice at days 1, 7, and 14 after ischemia-reperfusion injury (IRI), with or without Entospletinib treatment. Single-cell suspensions were pooled from 3 mice per condition. Live immune cells were enriched by FACS, fixed, and processed using the Evercode™ WT kit (Parse Biosciences) for single-cell RNA sequencing.
