Using the CRISPR genome editing system and a cancer cell/activated CD8+ T cell co-culture system, we identified the key genes in HCC cells that enhanced HCC cell tolerance to CD8+ T cells. Overall design: We performed a pooled CRISPR genome-wide loss/gain-of-function screen using a lentiviral KO/activation library in PLC-PRF-5/Hep-G2 cells. The cell pools were co-cultured with activated CD8+ T cells for 7 days
