The intermuscular bones (IBs) are attracting increasing attention considering its adverse effect on the edibleness and economic value of fish. Although a draft genome of Megalobrama amblycephala has been published, the completed sequences and structure of mRNA transcripts remain unclear, which limit further study on IB of that. In this study, we performed a PacBio Iso-Seq analysis for the mixed IB samples from 6- and 30-month-old M. amblycephala to explore the comprehensive genetic information including full-length transcripts, AS isoforms, APA, transcription factors, fusion genes, lncRNA, etc. The produced results not only revealed the full-length sequences and gene structure characteristics for IBs sample of juvenile and adult M. amblycephala, but also complement the genomic sequence and annotation information and provide important reference information for other fish research.
