The goal of the project was to conduct a transcriptome-wide study of the effects of actinomycin D (ActD) on gene expression and specific splicing events in a HeLa cell model of myotonic dystrophy. Actinomycin D decreases levels of the toxic CUG repeat containing RNA, likely through binding to the transfected CTG plasmid and disruption of transcription. Off target effects and specificity of Act D was investigated using RNA seq of the myotonic dystrophy model for Act D at levels: 0, 5nM, and 20 nM in duplicate.
