Community coalescence of microeukaryotes

Two microbiomes (river and sea) for a microcosms experiment were collected from a coastal area of the Gulf of Bothnia, Sweden. A 16-day long experiment were conducted, exposing river and sea communities (60 mL each) to one-time and repeated coalescences by applying three mixing ratios (1:1, 1:2 and 2:1, resp.). Amplification was done by using the V4_Balzano_F/D11_3143R primer pair in order to amplify a ~4.5 kbp region of the eukaryotic rRNA operon. Sequencing was performed using a MinION Mk1C instrument (ONT) operated with a Spot-ON Flow Cell (R9.4.1 chemistry). Real-time high-accuracy basecalling (HAC) was executed using the MinKNOW software (v22.05.6). The submitted raw sequences contain quality-filtered (Q > 9) reads.
