This data set contains concentrations of phytoplankton, protozoa, total bacteria and metabolically active bacteria assessed by flow cytometry on transects 12, 1, 3, 5, 7, 9 and 11 of the BROKE-West survey of the Southern Ocean between January and March 2006. Only total bacterial concentrations were assessed for transect 11. Between 4 and 12 depths were sampled for marine microbes and concentrations were assessed using FACScan flowcytometer. Phytoplankton were identified and counted based on the autofluorescense of chlorophyll a when excited by the 488 nm laser of the FACScan. Protozoa were identified and counted after staining with the acid vacuole stain Lysotracker Green. Total bacteria were identified and counted using the cell permeant SYTO 13 nucleic stain. 本数据集包含2006年1月至3月期间,在南大洋BROKE-West科考航次的断面12、1、3、5、7、9和11上,通过流式细胞术(flow cytometry)测定的浮游植物、原生动物、总细菌与代谢活性细菌浓度。其中仅断面11测定了总细菌浓度。本次科考针对海洋微生物共采集4至12个深度的样品,使用FACScan流式细胞仪完成浓度测定。浮游植物依据FACScan的488 nm激光激发叶绿素a产生的自发荧光进行识别与计数;原生动物经酸性液泡染色剂Lysotracker Green染色后完成识别与计数;总细菌采用细胞渗透性核酸染色剂SYTO 13进行染色,以此识别并计数;代谢活性细菌则通过酯酶染色剂6CFDA对胞内酯酶进行染色后完成识别与计数。本数据集包含以下字段:纬度、经度、断面编号、CTD编号、流式文件、深度(米)、总细菌(个/毫升)、活性细菌(个/毫升)、死亡细菌(个/毫升)、原生动物(个/毫升)、浮游植物(个/毫升)。本研究作为ASAC项目40(ASAC_40)的一部分完成。
