3D RNA-scaffolded wireframe origami

This study develops the first method of designing 3D wireframe nucleic acid origami with arbitrary RNA scaffolds, DNA staples, and double crossover edges. The 3D structure of the origami is solved using cryo-EM, and for the first time, the effects of sequence and structural features on stability of the origami are characterized at single-nucleotide resolution using DMS-MaPseq. These results indicate that arbitrary RNA scaffolds including mRNA can be folded into a variety of 3D geometries, with potential for developing these origami objects into nucleic acid vectors or therapeutics.
