We placed immotile variants of the soil bacterium Pseudomonas fluorescens Pf0-1 (Pf0-1(delta)fleQ; Pf0-2x) under selection for recovered flagella-mediated motility using soft agar as a directional selection medium. We observed that typically a single de novo mutation would arise within 72-144 hours and restore the motility phenotype. We sent a subset of evolved lines from this experiment for whole-genome sequencing to identify the phenotype-causing mutation and screen for secondary, background and hitchhiking mutations. One mutant line was identified to harbour a single nucleotide polymorphism within the glnA locus (glnA C5A), which is a member of the nitrogen regulatory pathway associated with recovery of the flagella-mediated motility phenotype.
