Epi-Decoder was used to delineate the proteome of a single tRNA gene (tP(UGG)M) in Saccharomyces cerevisiae to uncover novel regulatory mechanisms of RNAPIII. Comparing the proteome under different metabolic conditions (glucose vs glycerol 37C 2h) revealed reprogramming of the core RNAPIII transcription machinery. In addition, under conditions of nutrient stress we observed increased binding of heat shock factors, RNA processing factors, chromatin remodeling complexes and an uncharacterized factor Ykr011c/Fpt1. Deletion of FPT1 compromised the nutrient-dependent reprogramming of RNAPIII.
