LC variable region sequences from AL amyloidosis

Background: amyloid immunoglobulin light chain (AL) amyloidosis is caused by the misfolding of immunoglobulin light chains (IG LCs). Aim of this study was to validate the laser microdissection combining mass spectrometry (LMD/MS) method to identify the IGVL gene usage. Objective: We analyzed the LC variable sequence obtained by RNA-based immunoglobulin sequencing and LMD/MS analysis in ten AL patients. Material and Methods: RNA-based immunoglobulin sequencing and LMD/MS analysis in ten AL patients. Results: By applying RNA-based immunoglobulin sequencing, we successfully identified the dominant clonotypes in ten AL patients; LMD/MS analyais based on renal amyloid tissues recovered the IGVL region sequence and confirmed the specific IGVL gene in all cases. Conclusion: This study validated the LMD/MS methodology we applied.
