TNF synergy with IL-17A & IL-17F stimulation of MSCs

The IL-17 pathway is implicated in axial SpA and targeted by multiple biologics in the clinic, we wanted to assess the downstream effects of IL-17A and IL-17F on entheseal stroma as these cytokines have been identified to drive inflammation and possibly new bone formation. Specifically we wanted to investigate the synergistic effect of IL-17A and IL-17F in combination with TNF.
Bulk-seq was performed on spinal MSCs isolated from human spinal entheseal bone and soft tissue, MSCs were stimulated with IL-17A, IL-17F and TNF alone and in combination to assess the synergistic effects of these cytokines on stromal function and activation.
