Starting in late summer 2023, we decided to sequence a Romanian local breed, more specifically Baneasa 1 (B1) for the first time. The breed was provided by the Global Center of Excellence for Advanced Research in Sericulture and Promotion of Silk Production, University of Agricultural Sciences and Veterinary Medicine of Cluj-Napoca. Using genomic DNA extracted from B1 larvae, in January 2024 we made use of ONT sequencing and employed a MinION device in concert with the Ligation Sequencing Kit V14 methodology (along with the corresponding NEBNext-Companion Module for ONT) and R10.4.1 flow cells. Super accurate base-calling of POD5 sequencing output was performed with Dorado and FASTQ files were generated. The q7 filtered reads were submitted to de novo assembly using canu 2.2.
