CpG methylation analysis in Mesial temporal lobe epilepsy and Focal cortical dysplasia patients

CpG methylation analysis of MeDIP DNA using Agilent Human DNA methylation Microarray slides (G4495A, AMADID 023795) Using methylated DNA immunoprecipitation microarray (MeDIP-chip) and Agilent Human DNA methylation Microarray slides (G4495A, AMADID 023795) we report genomic methylation signatures of tissues resected from Mesial temporal epilepsy (MTLE) and Focal cortical dysplasia (FCD) type II patients undergoing surgery. Control samples were obtained from the non-epileptic post mortem cases without any brain pathology Tissues resected from 2 autopsy, 4 MTLE and 3 FCD type II were used for isolating Genomic DNA. Genomic DNA was extracted using genomic DNA extraction kit (Qiagen, Hilden, Germany), followed by MeDIP using monoclonal mouse anti 5-methylcytidine antibody. Both the Input and MeDIP DNA was amplified using GenomePlex Complete Whole Genome Amplification (WGA) kit. WGA amplified sample (MeDIP-WGA), along with their corresponding whole genomic DNA (input), were labeled usingthe SureTag DNA labeling kit (5190-3400, Agilent)and hybridized to Human DNA methylation Microarray slides (G4495A, AMADID 023795)
